Biglycan Antibody (3E2) is a mouse monoclonal IgG2a kappa light chain antibody that detects Biglycan protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-Biglycan antibody (3E2) is available as the non-conjugated form. Biglycan, a class I small leucine-rich proteoglycan (SLRP) found in the extracellular matrix, plays a crucial role in regulating bone cell differentiation and proliferation, which is vital for maintaining skeletal integrity and function. Biglycan is primarily located in the extracellular matrix, where Biglycan interacts with various growth factors and cytokines, thereby influencing cellular signaling pathways that govern bone metabolism. Notably, Biglycan contains two chondroitin sulfate glucosaminoglycan (GAG) chains attached near its amino terminus, distinguishing Biglycan from the closely related SLRP, decorin, which has only one GAG chain. This structural difference is significant as Biglycan′s ability to bind to collagen and other matrix components ultimately impacts tissue organization and repair. Studies have shown that Biglycan-deficient specimens exhibit a reduced capacity to generate bone cell precursors, a diminished response to transforming growth factor beta, decreased collagen synthesis, and increased apoptosis. In clinical contexts, patients with rheumatoid arthritis demonstrate heightened immune responses to Biglycan, while those with osteoarthritis do not, indicating that variations in immune recognition of SLRPs may contribute to the pathogenesis of inflammatory rheumatic diseases.

Which human tissue is it possible to use as positive control from paraffin embedded sections inorder to perform immunohistochemistry ?

Thank you for your question. Positive Controls for western blots are Biglycan (h2): 293T Lysate: sc-128099 or Hep G2 cell lysate: sc-2227. This particular protein has low tissue specificity, but can be found in connective tissues, especially in articular cartilages.

Can the Anti-Biglycan Antibody (3E2): sc-100857 be used for IHC? We have been using this antibody for WB of osteosarcoma cell extracts and IP (doi: 10.3390/cancers14051196.) and we want to try it for the histology of osteosarcoma biopsies. Thank you!

Thank you for your question. Anti-Biglycan Antibody (3E2): sc-100857 is only recommended for detection of precursor and mature Biglycan of mouse, rat and human origin by WB, IP and ELISA. Based on our tests with this product we do not recommend it for use in immunohistochemistry.
