    SEE ALSO...

BCAP Antibody (E-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects BCAP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). BCAP (E-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. The B cell adaptor for phosphoinositide 3-kinase (BCAP) plays a crucial role in B cell signaling by acting as a bridge between B cell receptor-associated kinases and the phosphoinositide 3-kinase (PIK3) pathway, which is essential for B cell activation and survival. Specifically, BCAP is phosphorylated by Syk, Btk, or Lyn kinases, creating binding sites for the p85 regulatory subunit of PIK3, thereby facilitating downstream signaling that influences cell proliferation and differentiation. BCAP mRNA expression is notably found in mouse spleen, thymus, liver, lung, macrophages, and B cell lines, highlighting its importance in immune responses. In humans, BCAP is located on chromosome 10q24.2, underscoring its evolutionary conservation and significance across species.

I see that there tends to be two predominant bands approximately between 90-55 K. Do these two bands represent different forms of the protein? What can I infer if I see one but not the other in a Western?

    Asked by: BenD
Thank you for your question. There are 3 predicted isoforms of BCAP described per Uniprot as 90 kDa, 70 kDa, and 45 kDa, respectively. Literature also describes the observance of multiple bands for BCAP due to alternative splicing of mRNA transcripts. You can find the sequences on Uniprot using the gene identifier Q6ZUJ8.

What blocking solution should I use with BCAP (E-5): sc-515498 monoclonal antibody for Western blot?

    Asked by: cjMara
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335). Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
