Anti-ATP6AP1 antibody (85.1) is available in a non-conjugated form, specifically designed for researchers seeking to study the vacuolar-type H+-ATPase (V-ATPase) complex. V-ATPase plays a crucial role in the acidification of eukaryotic intracellular organelles, which is essential for various cellular processes, including protein degradation, nutrient storage, and pH regulation. The V-ATPase is composed of two main domains: the peripheral V1 domain, responsible for ATP hydrolysis, and the integral V0 domain, which facilitates proton translocation across membranes. The V1 domain consists of nine subunits (A-H), while the V0 domain is made up of five subunits (a, d, c, c′, and c"). ATP6AP1, also known as 16A, CF2, Ac45, XAP3, ATP6S1, and VATPS1, is a type I transmembrane accessory protein predominantly expressed in endocrine and neuronal cells. ATP6AP1 plays a vital role in targeting the V-ATPase enzyme to specialized vacuolar systems, and through its cytoplasmic tail, ATP6AP1 interacts with the V0 domain subunits. Disruption of this interaction in osteoclasts has been shown to impair bone resorption, highlighting ATP6AP1′s importance in maintaining proper osteoclastic function and bone health.
