ASPH Antibody (A-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects ASPH protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-ASPH antibody (A-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Aspartyl/asparaginyl beta-hydroxylase (ASPH) is a crucial type II membrane protein primarily located in the endoplasmic reticulum, where ASPH plays a significant role in calcium homeostasis. ASPH is responsible for hydroxylating aspartate or asparagine residues in the epidermal growth factor-like (EGF) domains of several target proteins, utilizing iron as a cofactor in this enzymatic process. This post-translational modification is vital for proper functioning of these proteins, influencing their stability, activity, and interactions with other cellular components. ASPH gene encodes three distinct proteins—ASPH, Junctin, and Junctate (or Humbug)—which exhibit significant variation in their C-terminal domains and are expressed as five transcript variants. These variants differ in their roles related to calcium storage and release, hydroxylation capabilities, and tissue specificity, with all variants being present in skeletal muscle, while only some are found in heart, brain, pancreas, placenta, lung, liver, and kidney tissues. In the endoplasmic reticulum lumen, ASPH can be processed into two different forms, further underscoring functional diversity and importance in cellular processes.

We used this antibody in a Western Blot to see Junctin, but this was possible. What would you recommend me?

I am using ASPH (A-10): sc-271391 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
