ARK-1 Antibody (A-11) is a mouse monoclonal IgG2a antibody that detects Aurora related kinase-1 (ARK-1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ARK-1 plays a crucial role in cell division by regulating centrosome separation, bipolar spindle assembly, and chromosome segregation during mitosis, making ARK-1 essential for maintaining genomic stability. ARK-1 is primarily localized in the nucleus and associates with key components of the mitotic machinery, particularly at the centrosome and spindle poles, where ARK-1 is vital for proper mitotic progression. Notably, ARK-1 expression is significantly elevated in various colon carcinoma cell lines, indicating ARK-1′s potential involvement in oncogenesis. Furthermore, ARK-1 protein levels are tightly regulated, as ARK-1 accumulates during metaphase and is subsequently degraded after the G2/M transition via the ubiquitin-proteasome pathway, highlighting the importance of precise regulation in the cell cycle. The unique genetic loci of ARK-1 relative to ARK-2 suggests these kinases may have distinct yet complementary roles in cell cycle dynamics, further underscoring ARK-1′s significance in cancer biology and cellular function.
