ARC Antibody (G-4) is a mouse monoclonal IgG2b antibody that detects ARC in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ARC, also known as apoptosis repressor with CARD domain or nucleolar protein 3 (NOL3, NOP, NOP30), plays a crucial role in regulating apoptosis by inhibiting caspases, specifically caspase-2 and caspase-8, which are key mediators of programmed cell death. ARC′s ability to prevent apoptosis is vital for maintaining cellular homeostasis and survival, particularly in tissues like skeletal muscle and heart, where ARC expression is critical for cell viability under stress conditions. Notably, ARC is phosphorylated at Threonine 149 by casein kinase 2 (CK2), a modification that enhances anti-apoptotic function and facilitates mitochondrial localization, where ARC can effectively interact with caspase-8. This localization allows ARC to directly inhibit apoptosis triggered by various death receptors, including CD95/FAS and tumor necrosis factor receptor-1, thereby playing a protective role in cellular responses to stress and injury. With multifaceted functions and critical interactions, ARC is an important protein in the study of apoptosis and cellular survival mechanisms, making anti-ARC antibody (G-4) an essential tool for researchers investigating these pathways.
