API5 Antibody (E-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects API5 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). API5 (E-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. API5, also known as apoptosis inhibitor 5, AAC11, FIF, MIG8, XAGL, or API5L1, is a crucial antiapoptotic nuclear protein that plays a significant role in cellular survival and proliferation, making API5 a key player in cancer biology. API5 structure includes a nuclear localization sequence, an LxxLL motif, a leucine zipper domain, and a transactivation domain flanked by two acidic domains, which are essential for API5 function in regulating gene expression. Nuclear localization of API5 allows interaction with various transcription factors and signaling molecules, including forming a complex with fibroblast growth factor 2 (FGF-2), thereby mediating FGF-2-dependent signaling pathways. This interaction is vital for the regulation of matrix metalloproteinase-2 synthesis and the suppression of E2F-dependent apoptosis, linking API5 to tumor progression and malignancy in various cancer cell lines. API5 ability to modulate apoptotic pathways underscores API5 importance in cancer research and therapeutic development, making anti-API5 antibody (E-12) an invaluable tool for studying API5 role in cellular processes and disease mechanisms.
