SEE ALSO...

Apg-1 Antibody (D-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects Apg-1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Apg-1 (D-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Apg-1, also known as HSPA4L (heat shock 70 kDa protein 4-like) or Osp94 (osmotic stress protein 94), is a member of the heat shock protein (HSP) family, which plays a crucial role in cellular stress responses. Specifically, Apg-1 regulates protein folding and prevents protein aggregation, which is vital for maintaining cellular homeostasis, especially under stress conditions such as heat shock. Apg-1 forms an 839 amino acid homodimer that primarily localizes to the cytoplasm and nucleus, with the potential to translocate to the nucleus in response to stress, highlighting its importance in cellular signaling and stress response mechanisms. Apg-1′s ability to interact with other proteins and assist in their proper folding underscores its significance in various biological processes, making Apg-1 a valuable target for research in cellular biology and disease mechanisms.

I would like to know if this antibody has been tested for cross-reaction with Apg-2 (HSPA4/HSPH2) of HSP105 (HSPH1).

Asked by: Desp
Thank you for your question. We have not specifically tested our mouse monoclonal, Apg-1 Antibody (D-12): sc-133253, for cross-reactivity with Apg-2 or HSP105.

I am using Apg-1 (D-12): sc-133253 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Dr Ninau Qelp
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 1 out of 5 by from It recognised Apg-2 instead of Apg-1 on WBI tested it after siRNA treatment in HEK293 cells and after overexpression of V5-tagged Apg-1 and Apg-2. It failed to recognise Apg-1 and instead it recognised Apg-2 (upper band, between 100 and 130kD).

Rated 5 out of 5 by from Excellent antibody for Western blotting. This product has been independently tested and validated in-house at Kinexus Bioinformatics Corporation as a very potent and highly specific antibody for Western blotting with a mixture of mouse brain, testes and thymus tissue lysates. It strongly detects an appropriately sized target protein in immunoblots.
