AIM2 Antibody (B-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects AIM2 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). AIM2 (B-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AIM2, also known as Interferon-Inducible Protein AIM2 or PYHIN4, is a 343 amino acid protein belonging to the HIN-200 family and plays a crucial role in the immune response by detecting cytosolic DNA, which indicates infection or cellular damage. Upon binding to DNA, AIM2 forms a complex with the adaptor protein ASC, leading to Caspase-1 activation and pro-inflammatory cytokine release, thereby promoting an inflammatory response. This function is particularly important in tumor suppression, as AIM2 represses NFκB transcriptional activity, which is often upregulated in various cancers. Localized primarily in the nucleus, AIM2 contains one DAPIN domain and one HIN-200 domain, with the DAPIN domain facilitating protein-protein interactions and the HIN-200 domain enabling direct DNA binding. AIM2 is expressed at high levels in tissues such as the small intestine, testis, peripheral blood leukocytes, and spleen, and defects in AIM2 have been implicated in microsatellite unstable colon cancers, highlighting its significance in both immune regulation and cancer biology.

What is the recommended fixation method for immunofluorescence staining with AIM2 (B-8): sc-515514 monoclonal antibody?

Rated 1 out of 5 by from specificity for AIM2 is doubtfulWe have tested the AIM2 antibody (sc-515514) in WB on cell lysates upon stimulation with IFN-gamma and the AIM2 inducer poly-dA:dT, upon transfection with AIM2cDNA and upon transfection with siAIM2 (sc-88166). The detected signal does not differ between the lysates, although expression of AIM2 mRNA, that was analyzed in parallel from the same lysates, varies up to 1000 fold. The EGFP-AIM2 that should be detected as a signal of 65 kD was not detected at all. We therefore seriously doubt on the specificity of this antibody for AIM2!!
