AF-10 Antibody (HAF10 9A5/2) is a mouse monoclonal IgG2b kappa light chain antibody that detects AF-10 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). AF-10 (HAF10 9A5/2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AF-10 protein is a crucial nuclear transcription factor that plays a significant role in gene expression regulation, particularly in acute myeloid leukemia, where AF-10 is implicated in the t(10;11) chromosomal translocation. This translocation leads to fusion proteins that can disrupt normal cellular functions, making AF-10 a key player in oncogenesis. Human AF-10 protein consists of 1,027 amino acids and shares 90% identity with mouse AF-10, which contains 1,061 amino acids. AF-10 is predominantly expressed in testis, highlighting AF-10′s potential role in reproductive biology and development. AF-10′s ability to interact with other transcription factors, such as AF-17, further underscores AF-10′s importance in cellular signaling pathways and gene regulation, making AF-10 a valuable target for research in cancer biology and developmental studies.

How can AF-10 (HAF10 9A5/2): sc-53156 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: TinTin
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-53156 PE) or fluoroscein (sc-53156 FITC).
