Actin Antibody (SPM161) is a mouse monoclonal IgG1 kappa light chain antibody. Anti-Actin antibody (SPM161) is raised against Actin from the gizzard of chicken origin, exhibiting broad species reactivity including mouse, rat, human, avian, and canine in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP). Actin is a ubiquitous and highly conserved protein that can constitute up to 50% of the total protein content in eukaryotic cells, playing a pivotal role in various cellular functions such as muscle contraction, cell motility, and maintenance of cell shape and integrity. Actin filaments form both stable and dynamic structures within the cytoskeleton, serving as essential components of microvilli and the contractile apparatus of muscle cells. In higher eukaryotes, multiple isoforms of Actin exist, encoded by a family of genes and classified into three main types: α-Actin, primarily expressed in muscle tissues; β-Actin and γ-Actin, predominant in other cell types. The organization and dynamics of Actin filaments are regulated by small GTPase proteins like Rho, Rac, and Cdc42, which control the assembly of stress fibers, focal adhesions, and filopodia formation. Post-translational modifications of Actin, including phosphorylation, acetylation, and ubiquitination, are critical for modulating polymerization, interaction with Actin-binding proteins, and overall cytoskeletal dynamics, thereby influencing essential cellular processes like migration, division, and signal transduction. Dysregulation of Actin′s post-translational modifications can lead to various pathologies, highlighting the importance of studying Actin′s structure and function using Actin monoclonal antibody (SPM161).

I am using Actin (SPM161): sc-56459 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
