ACAD-8 Antibody (D-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects ACAD-8 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ACAD-8 (D-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ACAD-8, also known as Isobutyryl-CoA dehydrogenase (IBD) or activator-recruited cofactor 42 kDa component (ARC42), plays a crucial role in fatty acid and amino acid catabolism, providing essential energy for heart and skeletal muscle. Structurally, ACAD-8 consists of an N-terminal α-helical domain, a central β-sheet domain, and a C-terminal α-helical domain, which are vital for enzymatic function. This mitochondrial flavoprotein specifically participates in valine degradation, converting isobutyryl-CoA to methacrylyl-CoA, and localizes to mitochondrial matrix where ACAD-8 forms a homotetramer. ACAD-8 deficiencies can lead to serious health issues, including carnitine deficiency, dilated cardiomyopathy, and feeding intolerance in infants, with isobutyryl-glycine excretion in urine serving as a diagnostic marker for ACAD-8 related defects.
