



A20 Antibody (B-5) is a mouse monoclonal IgG1 antibody that detects A20 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). A20 is a Cys2/Cys2 zinc finger protein that plays a pivotal role in regulating inflammatory responses by acting as a negative regulator of nuclear factor κ B (NFκB) gene expression. This regulation is crucial because NFκB is a key transcription factor involved in the immune response, and uncontrolled activation can lead to chronic inflammation and various diseases. A20 is induced by inflammatory stimuli such as tumor necrosis factor (TNF) and interleukin 1 (IL-1), effectively terminating NFκB responses by inhibiting activation. A20′s structure includes seven zinc finger domains, with four being essential for in vitro inhibition of TNF-induced NFκB activation. A20 interacts with several proteins, including TRAF2, TRAF6, and IκB kinase (IKK) γ, which further enhances ability to prevent cell death. TXBP151, a novel A20-binding protein, may play a role in mediating A20′s anti-apoptotic activity. Given involvement in the negative feedback regulation of signal transduction, A20 and binding partners present promising avenues for therapeutic intervention in a range of inflammatory diseases.
