CD3 Antibody (HIT3a) is a mouse monoclonal IgG2a kappa light chain antibody, engineered to recognize the human CD3 and CD3/TCR complex, and demonstrates high reactivity in both immunofluorescence (IF) and flow cytometry (FCM) applications. CD3, also known as the T cell co-receptor complex or TCR-CD3 complex, is a vital component of the T cell receptor (TCR) signaling machinery located on the surface of T lymphocytes. Structurally, the CD3 complex comprises five invariant polypeptide chains, gamma, epsilon, delta, zeta, and eta, that assemble into three distinct dimers: γε, δε, and ζζ or ζη. This complex architecture is essential for transducing extracellular antigen recognition into intracellular activation signals, thereby facilitating T cell activation and proliferation. Each of the γ, ε, and δ chains contains a single immunoreceptor tyrosine-based activation motif (ITAM), while the ζ chain possesses three ITAMs, which amplify the signaling cascade upon antigen engagement. The precise arrangement and structural integrity of the CD3 complex enable the recruitment and activation of key protein kinases such as ZAP-70 and Syk, which are crucial for downstream signaling events. By targeting the CD3 complex, anti-CD3 antibody (HIT3a) allows researchers to meticulously study T cell activation, signal transduction pathways, and immune response mechanisms. Anti-CD3 antibody (HIT3a) accurately detects and analyzes CD3 expression on human cells, making CD3 (HIT3a) monoclonal antibody an indispensable tool for immunological research, disease modeling, and therapeutic development.
