Varicella Zoster Virus (VZV) glycoprotein IV (gpIV), also known as glycoprotein E (gE), is a vital component of the VZV, the virus responsible for chickenpox in primary infections and shingles (herpes zoster) in reactivated infections. gE is one of the most abundant and immunologically significant glycoproteins in the VZV envelope and plays a crucial role in the virus's life cycle, including cell-to-cell spread and viral pathogenesis.
gE is a transmembrane protein that is essential for the infectivity of VZV. It facilitates viral entry into host cells by binding to specific receptors on the cell surface, a key step in the initiation of infection. After the initial infection, the virus can remain dormant in nerve cells and can reactivate later in life, leading to shingles. During reactivation, gE is again critical for the spread of the virus from nerve cells to the skin, causing the characteristic rash and pain associated with shingles.Immunologically, gE is a major target of the host immune response. It elicits both humoral and cellular immune responses, making it a key antigen for vaccine development. The varicella vaccine, for instance, generates immunity against VZV partly by targeting gE and other viral components.
