SUN2 Activators

SUN2 Activators are a class of compounds that enhance the function of the SUN2 protein, a core component of the LINC complex that links the nucleoskeleton and cytoskeleton. They primarily work by influencing the structures and dynamics of the nuclear envelope, microtubules, and actin cytoskeleton, which SUN2 directly interacts with. Some of these activators, such as tamoxifen, affect the nuclearenvelope's structure, thereby enhancing SUN2's function. Others, like paclitaxel and nocodazole, interact with microtubules and indirectly enhance SUN2's functional activity. This is because SUN2 is involved in nuclear-cytoskeleton interactions, and changes in microtubule dynamics can have consequential effects on SUN2 function. Similarly, Cytochalasin D and Latrunculin A disrupt actin polymerization, which can also enhance SUN2 function due to its role in connecting the nuclear lamina to the actin cytoskeleton.

On the other hand, Diethylstilbestrol and Bisphenol A, synthetic compounds known for their interactions with estrogen receptors, modify the nuclear envelope structure. Since the nuclear envelope is the residence of SUN2, altering its structure can enhance the functional activity of SUN2. Lastly, compounds like Vinblastine, Vincristine, and Glyphosate disrupt microtubule formation, leading to an indirect enhancement of SUN2 function. By influencing the dynamics of the microtubules which are connected to the nucleus via SUN2, these compounds ultimately amplify the functional activity of SUN2.
