String Inhibitors

String inhibitors primarily target kinases involved in cell cycle regulation to modulate the activity and downstream effects of the String protein. For example, Roscovitine and Olomoucine are CDK inhibitors that act on CDK2 and CDK5, resulting in the inhibition of String-dependent cell cycle progression. This occurs because String is known to activate the CDKs, which are crucial for cell cycle advancement from G2 to M phase. PD0332991, another cell cycle inhibitor, specifically targets CDK4/6 and halts the phosphorylation of Rb, a substrate regulated by CDKs. By inhibiting CDK4/6, PD0332991 prevents Rb phosphorylation, resulting in cell cycle arrest and an overall reduction in the activation of String protein.

In contrast, Aurora kinase inhibitors like MLN8054, VX-680, ZM447439, and Hesperadin affect the role of String in different phases of mitosis. For example, MLN8054 inhibits Aurora A kinase, which is essential for entry into mitosis. This kinase functions downstream of String, and its inhibition effectively reduces String's role in mitotic entry. Hesperadin, an Aurora B inhibitor, narrows down its action to inhibit String's role in chromosome segregation during mitosis. Other kinase inhibitors like U0126, which targets MEK1/2, and SB203580, which inhibits p38 MAPK, disrupt String-dependent G2/M transition and stress response pathways respectively. By using these inhibitors, the specific biochemical roles that String plays in cell cycle progression and stress responses can be effectively modulated.
