SH3PXD2B Inhibitors

Chemical inhibitors of SH3PXD2B can exert their inhibitory effects through various mechanisms, primarily by targeting the Src family of tyrosine kinases, which are integral to the protein's function. Dasatinib, a potent tyrosine kinase inhibitor, can disrupt the interaction between SH3PXD2B and the Src kinases, thereby impeding the protein's role in podosome formation. Similarly, PP2 and SU6656, both selective Src family kinase inhibitors, are capable of obstructing the kinase activity essential for SH3PXD2B-mediated cytoskeletal rearrangements. Bosutinib and Saracatinib, also tyrosine kinase inhibitors, further contribute to the inhibition of SH3PXD2B by preventing the activation of Src kinases that SH3PXD2B requires for signaling pathways controlling cell structure and movement.

Furthermore, Herbimycin A, an ansamycin antibiotic, targets tyrosine kinases and consequently can hamper SH3PXD2B's role in cytoskeletal organization. Nilotinib, though primarily targeted at BCR-ABL kinases, can also inhibit the Src family kinases and thus indirectly inhibit SH3PXD2B. Ponatinib, another multi-targeted tyrosine kinase inhibitor, and KX2-391, a non-ATP competitive inhibitor of Src kinase, both can disrupt the kinase signaling pathways that are essential for SH3PXD2B's mediation of actin cytoskeleton dynamics. PP1, a selective inhibitor of Src family tyrosine kinases, and WH-4-023, which selectively inhibits Lck and Src family tyrosine kinases, can inhibit the SH3PXD2B by disrupting the Src kinase signaling, impairing the downstream processes SH3PXD2B is involved in within the cell, including cellular motility and invasion.
