SAP 61 Activators

SAP 61 Activators primarily function through influencing phosphorylation cascades and altering transcription regulations. For instance, Phorbol 12-Myristate 13-Acetate (PMA) is a potent activator of Protein Kinase C (PKC), a family of protein kinases involved in controlling the function of other proteins through direct phosphorylation. This activation of PKC can enhance the phosphorylation of the SF3A3 complex, of which SAP 61 is a part, and therefore increase SAP 61's splicing activity. Similarly, Calyculin A and Okadaic acidwork by inhibiting protein phosphatases 1 and 2A, leading to increased phosphorylation events in cells. This action can stimulate the functional activity of SAP 61 by enhancing the phosphorylation of the SF3A3 spliceosomal complex.

Another group of SAP 61 activators, including Forskolin, Etoposide, and Anisomycin, function by altering cellular signaling pathways. For instance, Forskolin activates adenylyl cyclase, which indirectly affects the PKA pathway. This alteration in the PKA pathway can boost the functional activity of SAP 61 by enhancing the phosphorylation of spliceosomal components like the SF3A3 complex. Etoposide, on the other hand, works by initiating the DNA damage response pathway, which can lead to an increase in splicing activity. This DNA damage response can indirectly enhance the function of SAP 61, a key player in the RNA splicing process. Anisomycin, a JNK activator, enhances the phosphorylation of the SF3A3 complex, leading to increased functional activity of SAP 61.
