RMP Inhibitors

Chemical inhibitors of RMP target various aspects of the cell cycle and mitosis, processes in which RMP is intricately involved. Alisertib, a known Aurora A kinase inhibitor, disrupts the cell cycle progression, and since RMP is associated with cell cycle regulation, its inhibition can lead to the indirect suppression of RMP's function. Similarly, VX-680 (Tozasertib) curtails Aurora kinase activity, which is crucial for mitotic spindle formation. Given RMP's role in cell cycle progression, the disruption of these kinases effectively undermines RMP's functionality. Roscovitine and PD 0332991, which selectively inhibit cyclin-dependent kinases (CDKs) and CDK4/6 respectively, arrest cell cycle progression. This arrest, specifically in the G1 phase by Palbociclib, hinders RMP's capacity to regulate the cell cycle and transcription, thereby inhibiting RMP. Dinaciclib, a potent inhibitor of various CDKs, further compounds this effect by targeting the CDKs that facilitate both cell cycle progression and transcriptional regulation, which are key areas of RMP involvement.

Continuing with the theme of cell cycle interference, AZD1152-HQPA and Barasertib, both selective inhibitors of Aurora B kinase, alter chromosome segregation and cytokinesis. RMP's role in these processes means that its functional activity is compromised when these kinases are inhibited. SNS-314, another Aurora kinase inhibitor, and ZM447439 follow a similar rationale; they disrupt mitosis, thereby indirectly impacting RMP's ability to regulate this process. BI 2536 takes a different approach by targeting Plk1, a protein essential for spindle assembly and cytokinesis, thus impeding RMP's associated functions. GSK1070916, which selectively inhibits Aurora B/C kinases, adds to this multi-pronged inhibition by disrupting cytokinesis and chromosome segregation. Finally, TAK-901, as a multi-kinase inhibitor, broadens this scope by affecting several kinases, including those that RMP interacts with, to regulate cell cycle progression and mitotic spindle assembly.
