Proliferin-1 Activators are a collection of chemical entities that serve to augment the functional activity of Proliferin-1 through multiple, distinct signaling cascades within the cellular milieu. The elevation of intracellular cyclic AMP levels by Forskolin and IBMX sets off a cascade of events, primarily through the activation of PKA, which is known to enhance the functional capacity of Proliferin-1 by phosphorylation of downstream targets. Epigallocatechin gallate and Genistein provide a different route to this enhancement by inhibiting competitive kinase pathways, which could allow Proliferin-1 associated signaling to become more prominent. PMA's role as a PKC activator further contributes to the upregulation of Proliferin-1 activity by promoting the phosphorylation of proteins within the same signaling pathways, while the lipid signaling mediator, Sphingosine-1-phosphate, asserts its effects by engaging its receptors to enhance related pathways.

Moreover, the manipulation of intracellular calcium levels by Thapsigargin and A23187 serves to activate calcium-dependent signaling routes, which hold substantial sway over Proliferin-1's activity landscape.
