PARP-4 Inhibitors

The chemical class of PARP-4 inhibitors primarily consists of compounds that are broad-spectrum inhibitors of the PARP enzyme family. These inhibitors function by binding to the PARP active site, preventing the enzyme from performing its role in DNA repair processes. This inhibition can lead to the accumulation of DNA damage in cells, which is particularly significant in cancer cells that rely on PARP-mediated repair mechanisms for survival. For example, inhibitors like Olaparib and Niraparib, initially developed for targeting PARP-1 and PARP-2, have shown potential in inhibiting other PARP family members, including PARP-4.

The relevance of targeting PARP-4 specifically is still an emerging area of research. While much of the focus has been on PARP-1 due to its prominent role in DNA repair, PARP-4 has distinct biological functions, including roles in chromatin structure regulation and gene expression. The inhibition of PARP-4, therefore, presents a unique opportunity to explore these specific cellular processes. However, the current limitation in the development of highly specific PARP-4 inhibitors necessitates the use of broader PARP inhibitors in research.
