Orai3 Inhibitors

Orai3 inhibitors refers to a class of chemicals that can reduce or block the activity of the Orai3 protein by interacting with the protein itself or affecting its associated pathways. These substances primarily operate by modulating cellular calcium dynamics. Chemicals such as Synta66 and GSK-7975A directly inhibit Orai1/Orai3 channels, blocking store-operated calcium entry (SOCE).

AnCoA4 functions by disrupting the interaction between Orai3 and STIM1, a crucial step in the activation of Orai channels. As a modulator of IP3 receptors, 2-APB can inhibit Orai channels at certain concentrations. Pyr6 is a selective inhibitor of Orai3 channels that suppresses SOCE. Compounds like SK&F 96365 and BTP2 inhibit receptor-mediated calcium entry and CRAC channels, respectively, impacting Orai3 function. Lastly, certain ions including Gadolinium (III) chloride, La3+, Ni2+, and Zn2+ are known to inhibit cationic channels, including Orai channels.
