Chemical activators of Olr851 can function through a variety of biochemical pathways to enhance its activity. Forskolin is one such activator, working by directly stimulating adenylate cyclase to increase the levels of cyclic AMP (cAMP) within the cell. The rise in cAMP subsequently activates protein kinase A (PKA), which can phosphorylate Olr851, leading to its activation. Similarly, 8-Bromo-cAMP, a cAMP analog, bypasses the adenylate cyclase step and directly activates PKA, which then proceeds to phosphorylate and activate Olr851. Ionomycin, through its ability to increase intracellular calcium levels, activates calcium/calmodulin-dependent protein kinases (CaMKs), which are then capable of phosphorylating Olr851. Phorbol 12-myristate 13-acetate (PMA) takes a different route by directly activating protein kinase C (PKC), which also targets Olr851 for phosphorylation and activation.
Further down the list of activators, Capsaicin works by triggering transient receptor potential vanilloid 1 (TRPV1) channels, leading to the activation of PKC and CaMKII, both of which are capable of targeting Olr851 for activation through phosphorylation. Calyculin A provides sustained activation by inhibiting protein phosphatases, which normally dephosphorylate and deactivate Olr851, thereby maintaining Olr851 in an active state. Anisomycin activates the c-Jun N-terminal kinase/stress-activated protein kinase (JNK/SAPK) pathway, which can result in the phosphorylation and subsequent activation of Olr851. Epidermal Growth Factor (EGF) activates the mitogen-activated protein kinase/extracellular signal-regulated kinase (MAPK/ERK) pathway, another avenue through which Olr851 can be phosphorylated and thus activated. Lithium Chloride inhibits glycogen synthase kinase-3 (GSK-3) within the Wnt signaling pathway, which indirectly leads to the activation of kinases that can phosphorylate Olr851, resulting in its activation. Hydrogen Peroxide induces oxidative stress pathways, which then activate kinases that can target Olr851, leading to its activation by phosphorylation. Isoquinolinesulfonamide modulates PKC activity, a key enzyme in the phosphorylation and activation of Olr851. Lastly, 1,9-Dideoxyforskolin acts on cellular kinases to phosphorylate and activate Olr851, yet uniquely does so without activating adenylate cyclase, showing a different mechanism of activation compared to Forskolin.
