MHC class II Inhibitors

MHC class II (Major Histocompatibility Complex class II) inhibitors represent a class of compounds designed to modulate the function of MHC class II molecules, which play a pivotal role in the immune system's adaptive response. MHC class II molecules are transmembrane proteins expressed primarily on the surface of antigen-presenting cells (APCs) such as dendritic cells, macrophages, and B cells. They are responsible for presenting exogenous antigens, typically from pathogens like bacteria and viruses, to CD4+ T cells. This interaction is fundamental for initiating the adaptive immune response and promoting T cell activation. MHC class II inhibitors function by interfering with various steps in this antigen presentation process. One primary mechanism by which MHC class II inhibitors operate is by targeting the transcriptional regulation of MHC class II genes. These genes encode the MHC class II molecules, and their expression is tightly regulated. MHC class II inhibitors can affect this regulation by influencing the activity of transcription factors like CIITA (Class II transactivator), which controls the expression of MHC class II genes. Additionally, inhibitors may interfere with the intracellular trafficking of MHC class II molecules or impair the loading of antigens onto these molecules within endosomal compartments. This can disrupt the proper assembly and presentation of antigen-MHC class II complexes, ultimately impeding the activation of CD4+ T cells and thereby modulating immune responses. MHC class II inhibitors encompass a diverse range of chemical entities, including small molecules and biologics, each with distinct mechanisms of action to regulate MHC class II function.
