Triptolide is known to exert its effects by inhibiting the activity of RNA polymerase II, which is essential for the transcription of mRNA. This inhibition can indirectly affect MED15 as it is part of the Mediator complex, which is crucial for RNA polymerase II-mediated transcription. Flavopiridol inhibits CDK9, a component of the positive transcription elongation factor b (P-TEFb), which is necessary for RNA polymerase II's transition from initiation to elongation.
