Ku70 Inhibitors

Ku70, also known as XRCC6, is a pivotal protein involved in DNA repair mechanisms, particularly in the non-homologous end joining (NHEJ) pathway. As a component of the Ku heterodimer alongside Ku80, Ku70 plays a crucial role in recognizing and binding to double-strand DNA breaks, thus initiating the repair process. The formation of the DNA-bound Ku heterodimer marks the initial step in the recruitment of downstream repair factors to the damaged DNA site, facilitating the accurate rejoining of broken DNA ends and ensuring genome stability. Additionally, Ku70's interaction with Ku80 forms the DNA-dependent protein kinase (DNA-PK) complex, which is essential for orchestrating NHEJ-mediated DNA repair. Inhibition of Ku70 disrupts the integrity of the DNA repair process, leading to genomic instability and increased vulnerability to DNA damage-induced cell death. Various mechanisms can mediate Ku70 inhibition, including interference with its interactions with other repair proteins, disruption of its DNA-binding ability, or inhibition of downstream signaling pathways crucial for DNA repair. These inhibition strategies impair Ku70's functionality, thereby compromising the repair of damaged DNA strands and rendering cancer cells more susceptible to the cytotoxic effects of DNA-damaging agents. Consequently, targeting Ku70 inhibition represents a promising approach for cancer therapy aimed at exploiting DNA repair vulnerabilities to selectively eliminate cancer cells while preserving normal cellular functions.
