KIAA0146, also known as Spatacsin or SPG11, is a protein that is encoded by the SPG11 gene in humans. This protein is implicated in a range of cellular functions, predominantly associated with the maintenance and organization of the endosomal-lysosomal system, which is crucial for the degradation and recycling of cellular components.The importance of KIAA0146 is underscored by its role in the pathogenesis of a form of hereditary spastic paraplegia (HSP), specifically spastic paraplegia type 11 (SPG11). HSP is a group of inherited neurological disorders characterized by progressive weakness and spasticity of the lower limbs, due to the degeneration of the corticospinal tracts. KIAA0146 is believed to be involved in the autophagic-lysosomal pathway, which is a system for degrading and recycling cytoplasmic components, including organelles, through the formation of autophagosomes that eventually fuse with lysosomes. Its role in neurodegeneration could be related to the impairment of this pathway, leading to the accumulation of undegraded material and subsequent neuronal damage.The protein's structure suggests that it may act as a scaffold, interacting with other proteins and possibly assisting in the assembly of multiprotein complexes that are important for autophagosome function.
