HSP 90α activators represent a diverse range of compounds that modulate the protein's activity through a variety of mechanisms. Some directly bind to HSP 90α, stabilizing it in an active form. Compounds like geldanamycin, radicicol, and 17-AAG are part of this subset. These compounds essentially capture HSP 90α in its activated state, promoting its chaperone functions. Others, such as novobiocin and purine, activate HSP 90α by affecting ATPase activity or by binding to the C-terminus of the protein. Another set of activators work by enhancing HSP 90α's interaction with client proteins. For example, celastrol promotes such interactions, thereby activating the chaperone activity of HSP 90α.
Some compounds, like staurosporine, function in a more indirect manner. Staurosporine does not interact directly with HSP 90α but elevates stress levels in the cell, which in turn enhances HSP 90α activity as part of the cellular stress response. Similarly, withaferin A upregulates the expression of HSP 90α. Other synthetic compounds such as SNX-2112 and BIIB021 have been designed to bind and stabilize HSP 90α.
