Chemical inhibitors of the Hedgehog pathway-related protein (HDRP) target various components of the Hedgehog (Hh) signaling cascade, which is crucial for the proper functioning of HDRP. Cyclopamine and Jervine are two such inhibitors that directly interact with Smoothened (SMO), a vital transmembrane protein in the Hh pathway. By binding to SMO, these molecules prevent its activation and subsequent signal transduction, effectively inhibiting the activity of HDRP. Other chemicals such as Vismodegib, LY2940680 (Taladegib), Sonidegib, Glasdegib, PF-5274857, and Itraconazole function similarly by inhibiting SMO, thereby blocking the downstream signaling required for HDRP's role in the pathway. These inhibitors ensure that the pathway remains inactive, which in turn means that HDRP does not carry out its function within the pathway.
In addition to SMO inhibitors, other chemicals target different aspects of the Hh pathway and related signaling mechanisms, thereby influencing HDRP activity. GANT61 disrupts the pathway at a later stage by antagonizing GLI transcription factors, which are downstream effectors in the Hh pathway. By preventing GLI from activating Hh target genes, GANT61 indirectly inhibits HDRP's function. TWS119 takes a different approach by inhibiting glycogen synthase kinase-3β (GSK-3β), a kinase involved in the Wnt signaling pathway, which intersects with Hh signaling and affects HDRP. Lastly, SAR245408 (XL147) inhibits phosphoinositide 3-kinase (PI3K), a kinase upstream in the AKT signaling pathway. Since HDRP is also influenced by AKT signaling, the inhibition of PI3K by SAR245408 leads to the suppression of HDRP's function within this context. All these chemical inhibitors collectively contribute to the regulation of HDRP by targeting various nodes and linkages within the complex network of cellular signaling pathways in which HDRP operates.

|---|---|---|---|---|---|---|
Cyclopamine | 4449-51-8 | sc-200929 sc-200929A | 1 mg 5 mg | $94.00 $208.00 | 19 | |
Cyclopamine specifically inhibits the Hedgehog signaling pathway by binding to and inhibiting Smoothened (SMO), a G protein-coupled receptor-like protein that is essential for the signal transduction. Since HDRP is involved in the Hedgehog pathway, the inhibition of SMO by Cyclopamine can lead to the downregulation of HDRP's activity. | ||||||
Vismodegib | 879085-55-9 | sc-396759 sc-396759A | 10 mg 25 mg | $82.00 $158.00 | 1 | |
Vismodegib directly binds to and inhibits Smoothened (SMO), thereby impeding the Hedgehog signaling pathway. This inhibition prevents the activation of downstream targets and transcription factors that are regulated by HDRP, thereby inhibiting HDRP's functional role in the pathway. | ||||||
GANT61 | 500579-04-4 | sc-202630 sc-202630A sc-202630B | 1 mg 5 mg 10 mg | $64.00 $131.00 $204.00 | 6 | |
GANT61 is a GLI antagonist that inhibits the Hedgehog signaling pathway by preventing GLI transcription factors from mediating the transcription of Hedgehog target genes. As HDRP's activity is linked to the Hedgehog pathway, the inhibition of GLI by GANT61 would suppress HDRP function. | ||||||
Jervine | 469-59-0 | sc-200934 sc-200934A | 1 mg 5 mg | $66.00 $240.00 | 1 | |
Jervine acts as an inhibitor of the Hedgehog signaling pathway by binding to Smoothened (SMO). This inhibitory action on SMO disrupts the pathway's function, which is necessary for HDRP activity, thereby leading to HDRP inhibition. | ||||||
Erismodegib | 956697-53-3 | sc-396280 sc-396280A | 10 mg 100 mg | $255.00 $918.00 | ||
Sonidegib is a selective inhibitor of Smoothened (SMO), a core component of the Hedgehog pathway. By inhibiting SMO, Sonidegib decreases the pathway's activity, thereby functionally inhibiting HDRP. | ||||||
Glasdegib | 1095173-27-5 | sc-507353 | 5 mg | $165.00 | ||
Glasdegib binds to and inhibits Smoothened (SMO), curbing the Hedgehog signaling cascade. This blockade prevents the proper functioning of the Hedgehog pathway, where HDRP operates, thus inhibiting HDRP's role. | ||||||
PF-5274857 | 1373615-35-0 | sc-478208 | 5 mg | $360.00 | ||
PF-5274857 is a potent and selective inhibitor of Smoothened (SMO), which is involved in the Hedgehog signaling pathway. By inhibiting SMO, it curtails the activation of HDRP-dependent processes within that pathway. | ||||||
Itraconazole | 84625-61-6 | sc-205724 sc-205724A | 50 mg 100 mg | $78.00 $142.00 | 23 | |
Itraconazole, though an antifungal agent, has been observed to inhibit Smoothened (SMO) and thus the Hedgehog signaling pathway. This inhibition affects the activity of HDRP as it is part of this pathway. | ||||||
