GLCE Inhibitors

Chemical inhibitors of GLCE can engage in various mechanisms to impede its function. Suramin, a polysulfonated naphthylurea, might inhibit GLCE by mimicking the structure of its natural substrates, which would block its function in heparan sulfate synthesis. Metal ions such as manganese(II) chloride, copper(II) sulfate, and zinc sulfate can disturb the metal ion balance required for GLCE's activity. Zinc sulfate may alter the enzyme's function by competing with the essential metal ions.

Further, sodium chlorate, as an oxidizing agent, can alter the redox state of cysteine residues within GLCE, potentially oxidizing critical thiol groups and thus impeding the enzyme's catalytic activity. Tunicamycin, which inhibits N-linked glycosylation, could indirectly inhibit GLCE by reducing the availability of properly glycosylated proteins necessary for its function. Sodium metavanadate can inhibit GLCE by mimicking the enzyme's phosphate group and binding to the active site, which would interfere with the enzyme's access to its substrate. Swainsonine, by inhibiting α-mannosidase II, can lead to a buildup of misprocessed glycoproteins, which could indirectly inhibit GLCE by disrupting its glycoprotein substrates or products. β-D-xyloside acts as a primer for glycosaminoglycan chains and competes with the substrates required by GLCE, thereby inhibiting its activity. Lastly, Rhodamine B, by non-specifically binding to the enzyme or its substrates, can lead to the inhibition of GLCE's enzymatic activity.
