Through its signaling pathways, GDF-6 plays a crucial role in regulating these processes. The activation of GDF-6 can be achieved by a diverse range of chemical compounds, each exerting its unique mechanism of action. One of the chemical activators of GDF-6 is SB431542, a selective inhibitor of the TGF-β type I receptor kinase. Other activators of GDF-6 include retinol, which binds to retinol-binding proteins and initiates gene expression changes; lithium chloride, which inhibits glycogen synthase kinase 3 (GSK-3) and enhances GDF-6 signaling; sodium butyrate, which inhibits histone deacetylases (HDACs) and leads to changes in gene expression; wortmannin, a potent inhibitor of phosphoinositide 3-kinase (PI3K) that indirectly activates GDF-6 by modulating downstream signaling pathways; genistein, a natural compound found in soybeans, that inhibits protein tyrosine kinases and modulates downstream signaling pathways; and curcumin, a natural compound found in turmeric, that modulates NF-κB and MAPK pathways, enhancing GDF-6 activity and impacting cellular responses.
