FBL8 Inhibitors

Chemical inhibitors of FBL8 function primarily through the mechanism of proteasome inhibition, which leads to an increase in cellular proteins that regulate the activity of FBL8. Proteasome inhibitors such as Velcade, Carfilzomib, Oprozomib, MLN9708, Marizomib, Epoxomicin, Lactacystin, MG132, and Delanzomib act to stabilize the intracellular proteins that are typically marked for degradation. This stabilization results in the accumulation of specific regulatory proteins that exert control over FBL8. As these proteins accumulate, they interact with FBL8, leading to a decrease in the functional activity of FBL8 by disrupting its normal regulatory processes. This disruption can inhibit the functional activity of FBL8 by preventing the normal proteolytic processing that FBL8 might require for its activity or by altering its substrate interactions. With the normal homeostasis of protein turnover being affected, FBL8's role within the cell is compromised, leading to its functional inhibition. Additionally, certain compounds like Aclacinomycin and Withaferin A, although not exclusively designed as proteasome inhibitors, have been found to inhibit the proteasomal pathway. Aclacinomycin does so by binding to the proteasome, which in turn can lead to the functional inhibition of FBL8 by similar mechanisms as those mentioned previously-specifically, by stabilizing proteins that regulate FBL8 and hinder its function. Withaferin A also exerts its effects through interference with the proteasomal pathway, resulting in the accumulation of regulatory proteins that interact with and inhibit the activity of FBL8.
