FAM173B Activators

FAM173B Activators work through a variety of molecular mechanisms to enhance the protein's functional activity. Some of these activators function by increasing the levels of intracellular cAMP, which in turn activates protein kinase A (PKA). The activated PKA can then phosphorylate FAM173B, resulting in its enhanced activity. This phosphorylation can occur through direct stimulation of adenylyl cyclase by certain compounds or through inhibition of phosphodiesterases (PDEs), which normally break down cAMP. By preventing cAMP degradation, these activators indirectly lead to higher PKA activity and subsequent phosphorylation of FAM173B. Additionally, agonists that stimulate G-protein-coupled receptors also increase cAMP levels, which similarly facilitates the activation of PKA. The consequence of this cascade is the phosphorylation and activation of FAM173B, aligning with the function of these activators.

Another set of activators exerts their influence through modulation of sirtuin activity. Sirtuins, which are NAD+-dependent deacetylases, can target FAM173B to deacetylate it, potentially leading to an increase in FAM173B's activity. Certain activators supply precursors that boost the intracellular levels of NAD+, thereby enhancing sirtuin activity and the subsequent deacetylation of FAM173B. Others, through their direct activation of sirtuins, can lead to similar outcomes.
