Dapper3 Inhibitors

Dapper3 inhibitors encompass a range of compounds primarily focused on the modulation of the Wnt/β-catenin signaling pathway. These compounds operate through various mechanisms to influence the pathway, indirectly affecting the activity of Dapper3. Dapper3, known for its role in Wnt signaling modulation, can be influenced by either stabilizing or promoting the degradation of pathway components, or by altering the secretion and activity of Wnt ligands themselves.

Some of these inhibitors target enzymes like GSK-3β, which when inhibited, can lead to the stabilization of β-catenin, thus diminishing the functional need for Dapper3's negative regulatory role. Others may inhibit proteins like tankyrase or porcupine, affecting the availability and activity of pathway components upstream of Dapper3. Additionally, inhibitors that block the interaction between β-catenin and TCF/LEF transcription factors also indirectly reduce the necessity for Dapper3's activity in the pathway.
