cornifelin Activators

Cornifelin (CNFN) is a protein associated with the cornified cell envelope of the epidermis, which is the outermost layer of the skin. It is encoded by the CNFN gene. Cornifelin is considered to be a late envelope protein, meaning it is one of the final components to be incorporated into the cornified envelope during the process of keratinocyte differentiation - the transformation of basal cells into corneocytes, which are the anucleated, dead cells that form the protective barrier of the skin.The presence of cornifelin is crucial for the proper assembly and function of the cornified envelope. This envelope is a structure of cross-linked proteins and lipids that provides a robust, protective barrier against environmental stressors such as pathogens, chemicals, and physical abrasion. By contributing to this barrier, cornifelin helps to maintain skin hydration and integrity.

Although cornifelin is not as well studied as some other proteins involved in epidermal barrier function, like filaggrin or loricrin, it is thought to have overlapping and unique roles in barrier formation and maintenance. Mutations or alterations in the expression of cornifelin may affect skin barrier function and could be implicated in skin disorders where the barrier is compromised.Given its role in the skin, cornifelin expression can be influenced by various factors that modulate keratinocyte differentiation and skin barrier homeostasis, including changes in humidity, UV radiation, and exposure to certain chemicals or irritants.
