Cornifelin (CNFN) is a protein associated with the cornified cell envelope of the epidermis, which is the outermost layer of the skin. It is encoded by the CNFN gene. Cornifelin is considered to be a late envelope protein, meaning it is one of the final components to be incorporated into the cornified envelope during the process of keratinocyte differentiation - the transformation of basal cells into corneocytes, which are the anucleated, dead cells that form the protective barrier of the skin.The presence of cornifelin is crucial for the proper assembly and function of the cornified envelope. This envelope is a structure of cross-linked proteins and lipids that provides a robust, protective barrier against environmental stressors such as pathogens, chemicals, and physical abrasion.
