Collapsin response mediator proteins (CRMPs) are a family of intracellular cytosolic phosphoproteins that play pivotal roles in regulating various cellular processes, primarily within the nervous system. CRMPs, also known as dihydropyrimidinase-related proteins, were initially discovered for their involvement in axon guidance, growth cone collapse, and neurite outgrowth during neuronal development.
CRMPs are known to be modulated through post-translational modifications, such as phosphorylation, and interactions with other proteins and ligands. Several signaling pathways and molecules indirectly influence CRMP function. For instance, Glycogen synthase kinase 3 beta (GSK-3β) is a kinase known to phosphorylate CRMPs, which can lead to the inhibition of their activity. Therefore, compounds that inhibit GSK-3β are considered potential modulators of CRMP activity, albeit indirectly. Similarly, various signaling cascades involving semaphorins, netrins, and other guidance cues have been found to regulate CRMPs. Researchers often investigate these pathways and their associated small molecules to gain insights into CRMP regulation.
