Carboxylesterase 3A Activators are chemical compounds that enhance the functional activity of carboxylesterase 3A through interactions that promote its enzymatic action. Bis(4-nitrophenyl) phosphate and Paraoxon, by acting as substrate mimics, can enhance the enzyme's hydrolytic capacity. Phenylmethylsulfonyl fluoride, at concentrations that do not inhibit the enzyme, may transiently enhance carboxylesterase 3A activity by inducing a beneficial conformational change. Similarly, Isoflurophate at low levels might transiently activate the enzyme through allosteric modulation before irreversible inhibition. Triton X-100 enhances substrate access to the enzyme, indirectly increasing its activity. Diisopropyl fluorophosphate and Carbaryl can interact with the active site, potentially leading to increased enzyme activity before irreversible inhibition or protection from other inhibitors, respectively.

Furthermore, Eserine can bind reversibly to carboxylesterase 3A and may enhance its catalytic efficiency by promoting optimal active site conformation. Malathion and Demeton-S-methyl, as organophosphate substrates, can enhance the enzyme's activity by increasing turnover rates. Methyl parathion, a similar substrate, can also be hydrolyzedefficiently by carboxylesterase 3A, potentially enhancing its functional activity. Lastly, Permethrin, through its interactions with carboxylesterase 3A, may serve as a readily hydrolyzable substrate, which would promote the enzyme's catalytic turnover, enhancing its activity.
