Cdc42EP4 activators are chemical compounds that enhance the functional activity of Cdc42EP4, a protein involved in cell morphogenesis and actin cytoskeleton organization. These activators work by influencing specific cellular processes and signaling pathways that Cdc42EP4 is directly involved in. Compounds such as EHT1864, Y-27632 and Fasudil enhance Cdc42EP4 activity by inhibiting other members of the Rho family of GTPases, such as Rac1 and ROCK. This inhibition leads to an increase in the relative activity of Cdc42, the protein that Cdc42EP4 directly interacts with, thereby indirectly enhancing the activity of Cdc42EP4.
Additionally, several Cdc42EP4 activators function by modulating the activity of Cdc42 and its downstream effectors. Wiskostatin, CK-666, CK-869, IPA-3 and FRAX597 inhibit the downstream effectors of Cdc42, including the N-WASP-Arp2/3 complex and PAK1. Inhibition of these effectors increases the availability of Cdc42, which in turn can enhanceCdc42EP4 activity.
