Ectonucleoside triphosphate diphosphohydrolase 6 (ENTPD6) is an enzyme that belongs to the family of ectonucleoside triphosphate diphosphohydrolases (ENTPDs), which are involved in the regulation of extracellular nucleotide levels. ENTPDs hydrolyze nucleoside triphosphates and diphosphates, such as ATP and ADP, to their monophosphate forms, and in some cases, further down to nucleosides. This hydrolytic activity plays a critical role in the termination of purinergic signaling, which is mediated by extracellular nucleotides acting as signaling molecules.

ENTPD6 specifically is distinguished by its preference for nucleoside diphosphates, such as UDP and GDP, over triphosphates like ATP. It is an integral membrane protein with its catalytic site facing the lumen of the endoplasmic reticulum (ER), suggesting a role in regulating nucleotide concentrations within the ER lumen, which can impact protein glycosylation and folding processes.The expression of ENTPD6 is tissue-specific, with significant levels found in the liver and placenta. Within cells, ENTPD6's localization to the ER suggests a specialized function in the maturation of glycoproteins, possibly by modulating the availability of UDP-sugars that are substrates for glycosyltransferases involved in glycosylation. Consequently, ENTPD6 may indirectly influence the stability and function of many secreted and membrane-bound glycoproteins.
