BUD31 Activators

BUD31 Activators are chemical compounds that enhance the functional activity of BUD31, a protein involved in the splicing of pre-mRNA within the spliceosome complex. Phosphatidic Acid, through the activation of MAPK, and Sphingosine-1-phosphate, by activating kinases that phosphorylate SR proteins, can enhance BUD31's activity in mRNA processing. These compounds work by promoting phosphorylation events that are essential for the regulation of splicing, where BUD31's role is crucial.

Compounds such as Ionomycin, Forskolin, and Bryostatin 1 manipulate intracellular signaling molecules like calcium, cAMP, and PKC, respectively, to influence the activity of BUD31. Ionomycin increases calcium levels, which may enhance the interaction of BUD31 with calcium-dependent splicing factors, while Forskolin raises cAMP levels, activating PKA which may indirectly enhance the functionof BUD31 by phosphorylating associated splicing factors. Moreover, Epigallocatechin gallate (EGCG) and Caffeine inhibit phosphodiesterases, increasing cAMP and cGMP levels, which activates PKA and PKG, leading to the phosphorylation of splicing factors and potentially enhancing BUD31 function in pre-mRNA splicing.
