ACTL7B Inhibitors

ACTL7B inhibitors primarily address actin dynamics or related signaling pathways, given the protein's connection to the actin family. One of the key targets is actin filament polymerization, which can be inhibited by Cytochalasin D, a compound that binds to the barbed ends of actin filaments, and Latrunculin A, which sequesters actin monomers. Contrarily, Phalloidin and Jasplakinolide stabilize actin filaments, with the latter even inducing their polymerization.

Further modulating actin dynamics, Swinholide A serves as an actin filament severing agent. On the regulatory front, CK-666, an Arp2/3 complex inhibitor, and SMIFH2, a formin inhibitor, both obstruct specific actin nucleation pathways. ML-141, Y-27632, and Wiskostatin target upstream regulators of actin polymerization, specifically CDC42, ROCK, and N-WASP, respectively. Myosin interaction with actin, another crucial component of actin dynamics, can be affected by Blebbistatin and BDM, both of which inhibit myosin ATPase activity, thereby modulating actin-myosin contractility.
