SphK1抗体(G-11)是一种IgG1 κ小鼠单克隆SphK1抗体(也称为SPHK1抗体、SK1抗体、SPHK抗体、SPK抗体或鞘氨醇激酶1抗体),可通过WB、IP、IF和ELISA检测小鼠、大鼠和人类来源的SphK1蛋白。SphK1抗体(G-11)既可作为非共轭抗SphK1抗体形式,也可作为多种共轭形式的抗SphK1抗体,包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®共轭物。鞘氨醇激酶(SphK或SphK1)是一种关键酶,催化鞘氨醇的磷酸化以形成鞘氨醇1-磷酸(SPP或S1P)。SPP是一种生物活性脂质,在多种细胞类型中发挥多种生物学效应,既可作为细胞内信使,也可作为与Edg家族受体结合的细胞外配体。SphK1的竞争性抑制剂可阻断SPP的形成,并选择性抑制由多种因素诱导的细胞增殖。SphK1活性的一个有效抑制剂是DMS(N,N-二甲基鞘氨醇)。SPP/SphK1已被认为是一种信号通路,可调节多种细胞功能,包括细胞生长、增殖和存活。具体来说,SphK1参与保护人肝细胞免受TNFα凋亡作用的信号通路。此外,SPP/SphK1可能通过调节SAPKs和caspases的激活在神经元存活中发挥重要作用。SphK1在成人肝脏、肾脏、心脏和骨骼肌中的表达水平最高,但SphK1的激活使细胞脱离其肝特异性表型。SphK1与SphK2高度同源,SphK2是一类不断增长的鞘脂激酶中的另一个成员。SphK2 mRNA的表达表现出与SphK1明显不同的组织分布,并出现在胚胎发育的较晚阶段。

Rated 4 out of 5 by from Specific for WBTested on SPHK1 K.O. together with control on Brain tissue. Showed no bands in the mutant. However, a bit dirty.

Rated 1 out of 5 by from Does not recognize SphK1While this antibody recognizes a band in the correct MW range of SphK1, it does not appear to actually be specific for SphK1. We tested the antibody by western blotting using wild type and SphK1 KO knockout cell lysate as well as with recombinant crystallography grade SphK1 as a positive control. Both cell lysates were immunopositive, but the positive control was not. If you would like me to upload images please let me know. We did receive this antibody as a free sample, and our lab has extensive experience with this protein.

Rated 5 out of 5 by from Potent and very specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was medium-strong detection of the ~42 kDa target protein in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested.
