Pit-1 Antibody (D-7) is a mouse monoclonal IgG2a kappa light chain antibody that detects Pit-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pit-1 (D-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pit-1, also known as growth hormone factor-1 (GHF-1), is a crucial transcriptional regulator that plays a significant role in the development of the anterior pituitary gland, where Pit-1 is essential for the differentiation and function of somatotropes, lactotropes, and thyrotropes. Pit-1 is characterized by a POU domain, which contains a bipartite DNA-binding domain vital for regulatory functions. Pit-1′s ability to activate growth hormone and prolactin transcription is critical for normal growth and metabolic processes, while Pit-1′s role as a repressor of gene expression facilitates specialization of various cell types within the pituitary. Pit-1 exists in two alternatively spliced forms, Pit-1a and Pit-1b, which exhibit different trans-activation capabilities, further underscoring regulatory complexity. Mutations in Pit-1 can lead to combined pituitary hormone deficiency (CPHD), affecting production of growth hormone, prolactin, and thyroid-stimulating hormone, highlighting Pit-1′s importance in endocrine health. Pit-1 is located on human chromosome 3p11, making Pit-1 a significant target for research in developmental biology and endocrinology.

hi, We have purchased D7 sc393943 por formalin fixed paraffin embedded tissues on Ventana benchmark ultra platform. Does it work? At what dilution? pretreatment etc? Thks

Thank you for your question. D7 antibody (sc-393943) has not been specifically validated for use on the Ventana Benchmark Ultra platform with formalin-fixed, paraffin-embedded (FFPE) tissues. However, customers have reported successful results with this antibody in immunohistochemistry (IHC) applications on FFPE samples.

Can this antibody be used in immunohistochemistry for formalin fixed paraffin embedded samples?

Thank you for your question. We have not yet tested this monoclonal antibody, Pit-1 (D-7): sc-393943, for formalin-fixed paraffin-embedded tissue sections and so IHC(P) is not a recommended application at this time.
