Keap1 Antibody (A-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects Keap1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Keap1 antibody (A-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Keap1, or kelch-like ECH-associated protein 1, functions as a stress-sensing adaptor for the Cullin3 (Cul3)-dependent E3 ubiquitin ligase complex, essential for regulating Nrf2 (NF-E2-related factor 2). The interaction between Keap1 and Nrf2 maintains cellular homeostasis, as Keap1 negatively regulates Nrf2 under normal conditions, preventing nuclear accumulation. Upon exposure to oxidative stress or electrophiles, Keap1 undergoes conformational changes that activate Nrf2, enabling nuclear translocation and expression of cytoprotective genes. This dynamic regulation supports cellular defense mechanisms against oxidative damage, underscoring Keap1′s role in maintaining cellular integrity and response to environmental stressors. The specific lysine residues targeted by Keap1 for ubiquitination are located in the N-terminal Neh2 domain of Nrf2, while cysteine residues Cys 273 and Cys 288 in Keap1 are crucial for suppressing Nrf2 activity.
