IL-6抗体（10E5）是一种IgG2b κ小鼠单克隆IL-6抗体（也被称为IL-6抗体或白细胞介素6抗体），它通过WB, IP, FCM和ELISA检测小鼠和大鼠来源的IL-6。IL-6抗体（10E5）以非偶联的抗IL-6抗体形式提供。白细胞介素-6，或IL-6，是一种多功能蛋白质，长度为212个氨基酸，它在宿主防御、免疫反应和造血中起着关键作用。IL-6由表皮朗格汉斯细胞组成型表达，并在受刺激的角质形成细胞中诱导表达。IL-6、IL-1β和TNFα作为内源性致热源，调节对细菌入侵的发热反应。IL-6受体是一个三聚体复合物，由IL-6特异性α链和IL-6、IL-11、CNTF、OSM和LIF受体共有的gp130糖蛋白的同源二聚体组成。IL-6的刺激导致gp130同源二聚化和相关激酶JAK1和JAK2的激活。一旦被激活，JAK1和JAK2会磷酸化Stat3，导致其核转位和Stat3响应基因的转录。此外，IL-6还被证明可以激活Ras/MAP激酶途径，该途径调节NFIL6转录。

Can we use this IL-6 (10E5) antibody (sc-57315) for IHC?

Thank you for your question. IL-6 Antibody (10E5): sc-57315 is only recommended for detection of IL-6 of mouse and rat origin by WB, IP, FCM and ELISA. We do not recommend it for use in IHC or IHC(P) applications.

In figure (from left to right): 30ug, 60ug, 80ug and 100ug of extract. The signal was slightly non-specific, but a more intense band was observed around 21kDa, as suggested by the datasheet. [Marker (from the bottom): 11,17,22,25,32 kDa]

Antigen retrieval methods were optimized between without antigen retrieval, heat-induced citrate buffer, and trypsin-induced antigen retrieval. Positive signals in portal tracts were clear without any antigen retrieval.
