FXR Antibody (D-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects FXR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-FXR antibody (D-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FXR, also known as NR1H4, plays a crucial role in regulating bile acid homeostasis and lipid metabolism, making FXR essential for maintaining metabolic health. Located primarily in the liver and intestine, FXR functions as a nuclear receptor that, upon binding to ligands, regulates the expression of genes involved in bile acid synthesis and transport. This regulation is vital because dysregulation of bile acids can lead to metabolic disorders, liver diseases, and other health issues. FXR forms heterodimers with retinoid X receptor (RXR) and binds to specific DNA sequences known as response elements, thereby influencing gene transcription. FXR′s ability to interact with various lipid molecules, including farnesoids, further underscores FXR′s importance in metabolic signaling pathways. With diverse applications and critical role in metabolic regulation, anti-FXR antibody (D-3) serves as an invaluable tool for researchers studying the complexities of lipid metabolism and related diseases.

Is it possible that we are seeing FXR at around 50kd rather than 75kd in the intestine using western blot analysis? I am aware from the data sheet that it says: Molecular Weight (predicted) 56 kDa. Molecular Weight (observed) 75 kDa. So i

thank you for your question. Isoform 5 of FXR (identifier: Q96RI1-5) maps at about 50 kDa, so it is possible that this is the isoform you are detecting in your samples.

Can this antibody be used in mouse cell line for detection of FXR by western blotting?

Thank you for your question. We haven’t tested this antibody yet in mouse cell lines, and so that is not a recommended species at this time. However, there are product citations and customer reviews indicating this antibody can be used successfully for detection of mouse FXR by Western blotting.

When should the TransCruz reagent sc-25309 X be used instead of the standard form sc-25309?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Hi, There is any publication where this antibody was used in mouse tissue. I'm interested in staining FXR in mouse intestine. Thanks in advance

Thank you for your question. We have not yet tested this antibody on mouse tissue. We are aware of a publication citing use of the antibody for Western blot of mouse liver samples. Please refer to the following reference on PubMed in Mol Cell Biol., PMID: 22290433. We have not yet tested this antibody for IHC on formalin-fixed paraffin-embedded tissue sections. It is currently recommended for IF, or immunofluorescence staining of cells or tissues on fresh/frozen tissue sections fixed in acetone or methanol. Rated 3 out of 5 by from 只能显细胞过转的FXR;显示不了组织中的,组织中相应的条带是杂带。显示不了组织中的,组织中相应的条带是杂带,用了FXRKO鼠的组织做阴性对照,也在相应未知出现条带,但是293T细胞过表达FXR时能显示出过量表达的FXR。 故最后结论:只能显细胞FXR,不能显示组织FXR!
