PDH-E1α Antibody (D-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects PDH-E1 alpha protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PDH-E1α (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The pyruvate dehydrogenase (PDH) complex, which includes the PDH-E1α subunit, is crucial for cellular metabolism as PDH-E1α serves as the primary link between glycolysis and the tricarboxylic acid (TCA) cycle by catalyzing the irreversible conversion of pyruvate into acetyl-CoA. PDH-E1α subunit is a heterotetramer composed of two alpha and two beta subunits, with the E1 active site located within the E1-α subunit, making PDH-E1α essential for the overall function of the PDH complex. PDH-E1α activity is tightly regulated through phosphorylation and dephosphorylation, which is vital for maintaining metabolic homeostasis. PDH-E1α gene is located on chromosome Xp22-q24, and mutations, such as a 20-bp deletion in the last exon, can lead to PDH deficiency, resulting in severe clinical manifestations including seizures, mental retardation, spasticity, and episodes of lactic acidosis associated with cerebellar ataxia.
