CTLA-4 Antibody (F-8) is a mouse monoclonal IgG1 kappa light chain antibody specifically engineered to target cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), also known as CD152, by recognizing an epitope within amino acids 195-223 at the C-terminus of the human protein. CTLA-4 is a crucial immune checkpoint receptor predominantly located on the surface of activated T cells, where CTLA-4 plays a vital role in downregulating immune responses by competing with the stimulatory receptor CD28 for binding to B7 ligands (CD80/CD86) on antigen-presenting cells. This strategic cellular localization is essential for maintaining immune homeostasis and preventing excessive immune activation that could lead to autoimmunity, making anti-CTLA-4 antibody (F-8) a key target for immunological research and therapeutic interventions. CTLA-4 monoclonal antibody (F-8) demonstrates broad species reactivity, effectively detecting CTLA-4 from mouse, rat, and human origins in various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA), as well as cross-reacting with CTLA-4 from equine, canine, bovine, and porcine species. CTLA-4 (F-8) antibody serves as an invaluable tool for investigating the inhibitory pathways that modulate T cell activation and proliferation, thereby advancing research in immunology, cancer immunotherapy, and autoimmune disease models. What is the specific region where this antibody is interacting? The datasheet says that the region of the interaction is between amino acids 195-223, but this is bigger region than an epitope region (6-8 aa). Is it recognizes a discontinuous epitope?

I have purchased the following antibody (CTLA-4 Antibody (F-8): sc-376016) to assess the ctla-4 expression with western blot on un-activated whole human PBMC. I have used 80,000 cells but couldn’t get a signal. Can you please suggest if anyone
