ZO-3 Antibody (1E8) is a mouse monoclonal IgG2a kappa light chain antibody that detects ZO-3 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-ZO-3 antibody (1E8) is available as the non-conjugated format. ZO-3 protein plays a crucial role in the formation and maintenance of tight junctions, which are essential for establishing and regulating the paracellular barrier in epithelial and endothelial cells. These tight junctions not only serve as physical barriers to prevent the passage of solutes and pathogens between cells but also play a significant role in cell signaling and maintaining cell polarity. ZO-3, along with other members of the membrane-associated guanylate kinase (MAGUK) family, such as ZO-1 and ZO-2, contributes to the structural integrity of these junctions through unique protein-protein interaction domains, including PDZ, SH3, and guanylate kinase domains. The interaction of ZO-3 with other tight junction proteins, such as occludin and claudins, is vital for proper assembly and function of tight junctions, which in turn influences various physiological processes, including nutrient absorption and immune response. Notably, alterations in ZO-3 expression or function have been implicated in various diseases, including cancer, where tight junction integrity is often compromised, leading to increased permeability and metastasis.
